The Invitrogen™ Human Cortisol Rapid ELISA Kit is a solid-phase, competitive enzyme-linked immunosorbent assay designed for the quantitative measurement of cortisol in various human biological matrices, including serum, plasma, urine, and saliva. The assay delivers high-throughput results with excellent sensitivity and precision, utilizing a 90-minute total assay time and requiring only one wash step, making it ideal for time-sensitive research environments.

The assay employs a cortisol-specific HRP-conjugated antibody, which competes with free cortisol in the sample for binding to immobilized cortisol on the microplate. The extent of binding is inversely proportional to the cortisol concentration. A colorimetric signal is developed using TMB substrate and measured at 450 nm, allowing for accurate quantification of cortisol levels in test samples.

    Key Features

    • Rapid format: 90-minute total assay time with only one wash step
    • Minimal hands-on time: Approximately 15 minutes
    • Validated for multiple sample types: Serum, plasma, saliva, and urine
    • High sensitivity: 2.87 ng/mL
    • Dynamic range: 6.25 to 400 ng/mL
    • Precision: Intra- and inter-assay CVs <10%
    • Readout: Colorimetric detection at 450 ± 2 nm
    • Sample volume required: 50 µL per well
    • Species specificity: Human

    Assay Principle

This is a competitive ELISA, where cortisol in the sample competes with a known amount of HRP-conjugated cortisol antibody for binding sites on the cortisol-coated microplate. Following a single wash step, TMB substrate is added to initiate a color reaction. The color intensity is inversely proportional to the cortisol concentration in the sample. Absorbance is read at 450 nm to determine quantitative results.

    Target Background

Cortisol is a key glucocorticoid hormone secreted by the adrenal cortex. It plays a central role in stress response, immune function, glucose metabolism, and circadian rhythm regulation. Its levels in body fluids are tightly regulated by the hypothalamic-pituitary-adrenal (HPA) axis through ACTH and CRF signaling. Quantifying cortisol levels is essential for understanding adrenal function and stress-related conditions.

    Applications

    • Stress response and circadian rhythm studies
    • Endocrinology and adrenal function research
    • Hormonal feedback loop analysis
    • Non-invasive cortisol monitoring using saliva or urine
    • Biomarker studies in immunology and metabolism

The Invitrogen™ Human Cortisol Rapid ELISA Kit offers a robust, user-friendly solution for the rapid quantification of cortisol in various human samples. With high sensitivity, excellent reproducibility, and compatibility with multiple matrices, this kit is ideally suited for research in endocrinology, stress biology, and metabolic regulation. Its streamlined protocol reduces hands-on time while maintaining assay integrity, making it a reliable tool for both routine and advanced laboratory applications.
