The Invitrogen™ Alanine Aminotransferase (ALT/GPT) Activity Kit is a sensitive and reliable colorimetric assay designed for the quantitative determination of ALT activity in a variety of biological samples, including serum, plasma, tissue homogenates, cultured cells, and cell culture supernatants.
Alanine aminotransferase (ALT/GPT) is a pyridoxal phosphate-dependent enzyme involved in amino acid metabolism and is widely used as a clinical biomarker for liver function, including conditions such as liver cirrhosis, viral hepatitis, and alcoholic liver disease.
This assay is based on the enzymatic conversion of alanine and α-ketoglutarate to pyruvic acid and glutamic acid, followed by formation of a colored phenylhydrazone complex, which is quantified spectrophotometrically at 510 nm.

Assay Principle
ALT catalyzes the conversion of:
- Alanine + α-ketoglutarate → Pyruvic acid + Glutamic acid
The generated pyruvic acid reacts with phenylhydrazine to form a reddish-brown phenylhydrazone complex under alkaline conditions. The intensity of the color, measured at 510 nm, is directly proportional to ALT activity in the sample.
