Sensitive fluorescence-based assay for quantifying lysozyme activity in solution

The Invitrogen™ EnzChek™ Lysozyme Assay Kit offers a fast, simple, and highly sensitive method to measure lysozyme activity in solution using a fluorescence-based readout. Capable of detecting lysozyme concentrations as low as 20 U/mL, the kit is compatible with any fluorescence microplate reader or fluorometer set to standard fluorescein (FITC) excitation/emission settings.

The assay utilizes fluorescein-labeled Micrococcus lysodeikticus cell walls in which the fluorescence is quenched until the substrate is hydrolyzed by lysozyme. The enzyme cleaves the β-(1→4)-glucosidic linkages between N-acetylmuramic acid and N-acetyl-D-glucosamine in the bacterial cell wall, relieving the quench and producing a dramatic fluorescence increase proportional to lysozyme activity.
