This broth serves as an alternative to Thioglycollate Medium USP CM0173, specifically designed for sterility testing of turbid or viscous biological products. The formulation excludes agar and resazurin, making it essential for freshly prepared or recently boiled medium usage.
Key Features
- Application:
- Used for sterility testing of turbid/viscous samples where regular thioglycollate media are unsuitable due to viscosity.
- Suitable for inactivating mercurial preservatives (up to 0.03% w/v) in inocula.
- Advantages:
- Lacks agar and resazurin, ensuring better adaptability for challenging biological products.
- Ideal for samples requiring freshly prepared or minimally oxidized media.
- Compatibility:
- Recommended for use with turbid or viscous inocula.
- Designed to meet standards outlined in N.I.H. Memorandum and USP XXI.
Typical Formula (gm/litre)
- L-cystine: 0.5
- Sodium chloride: 2.5
- Glucose: 5.5
- Yeast extract: 5.0
- Pancreatic digest of casein: 15.0
- Sodium thioglycollate: 0.5
- pH: 7.1 ± 0.2 @ 25°C
Preparation Instructions
- Dissolution: Suspend 29g of the medium in 1 litre of distilled water and bring to a boil until fully dissolved.
- Sterilization: Distribute the medium into tubes or bottles and autoclave at 121°C for 15 minutes.
- Usage: Prepare freshly or boil and cool the medium within four hours before use.
Storage & Shelf Life
- Dehydrated Medium: Store at 10–30°C.
- Prepared Medium: Not recommended for storage; can be held at 20–30°C in the dark for short periods.
- Avoid reheating the medium multiple times to prevent the formation of toxic oxygen radicals.
Appearance
- Dehydrated Medium: Straw-colored, free-flowing powder.
- Prepared Medium: Straw-colored solution.
Quality Control
Control Organisms | Expected Results |
---|---|
Candida albicans ATCC® 10231 | Flocculent growth |
Clostridium sporogenes ATCC® 19404 | Turbid growth and/or colonies |
Peptostreptococcus anaerobius ATCC® 27337 | Turbid growth and/or colonies |
Negative Control | No change (uninoculated medium) |
Precautions
- Use the medium freshly prepared or within four hours of preparation.
- Avoid reheating the medium multiple times to prevent oxidative damage.
- Ensure appropriate storage conditions to maintain efficacy.
