The Thermo Scientific™ Uracil-DNA Glycosylase, heat-labile (UDG/UNG), is a recombinant enzyme derived from Gadus morhua (cod liver). It excises uracil residues from DNA by cleaving the N-glycosidic bond between the uracil base and sugar backbone, generating apyrimidinic (abasic) sites that block replication and hybridization.

Unlike standard UNG enzymes, this heat-labile variant is completely and irreversibly inactivated after 10 minutes at 50 °C, making it ideal for workflows that require carry-over contamination prevention with subsequent PCR, cloning, or sequencing.

Key Features

  • Heat-Labile – Fully inactivated after 10 min at 50 °C; no risk of residual activity.
  • Specificity – Excises uracil from both single- and double-stranded DNA; inactive on RNA and dT-containing DNA.
  • Contamination Prevention – Eliminates uracil-containing DNA from prior PCR amplifications.
  • Enhanced Workflow Efficiency – Improves cloning success and site-directed mutagenesis efficiency.
  • Purity Guaranteed – Chromatographically purified and tested free from contaminating endonucleases and exonucleases.
  • Flexible Applications – Compatible with DNA repair, protein–DNA interaction studies, mutagenesis, and SNP detection.

Applications

  • PCR carry-over contamination prevention
  • Study of DNA repair and mutation detection
  • Increase cloning efficiency of PCR products with dU-primers
  • Site-directed mutagenesis enhancement
  • Protein–DNA interaction research
