How It Works:
- Sample Preparation:
- Homogenize a food sample in Buffered Peptone Water (BPW) or Ringer's Solution to create a 1:10 dilution.
- Incubate the sample suspension at 36 ± 1°C for 12-18 hours.
- System Inoculation:
- Add 0.5 mL of the sample suspension into the provided Physiological Solution.
- Distribute 0.2 mL of the suspension into each well.
- Incubation:
- Cover specific wells with Vaseline Oil (e.g., LDC, H2S, UR) and incubate at 36 ± 1°C for 18-24 hours.
- Result Interpretation:
- Observe color changes in the wells to detect and presumptively identify microorganisms.
- For example:
- Salmonella spp.: Yellow to red/orange (LDC) or yellow to black (H2S).
- E. coli: Red to blue (ESC) with pink-red after adding Kovac’s Reagent (IND).
- Listeria spp.: Yellow to black (LIS) and bubble formation in CAT after adding H2O2.
- Optional Confirmation:
- Perform biochemical or serological tests (e.g., oxidase test for Pseudomonas spp.) or plate onto selective agar for further confirmation.
