Principle of the Assay
This ELISA utilizes a double-antibody sandwich technique. The pre-coated microplate contains an anti-human OxLDL capture antibody. After sample addition, HRP-conjugated detection antibody binds to the OxLDL-antibody complex. Upon substrate addition (TMB), color develops proportionally to the amount of OxLDL. The reaction is stopped, and absorbance is read at 450 nm, allowing quantification based on a standard curve.

Sample Types & Preparation
Validated for:
- Serum & Plasma: Centrifuge at 2,000–3,000 rpm for 20 minutes.
- Cell Culture Supernatants: Centrifuge post-culture.
- Tissue Homogenates: Homogenize in PBS (pH 7.4) at 4 °C, centrifuge, and collect supernatant.
- Urine & Body Fluids: Same procedure as above.
Avoid sodium azide (NaN₃) and repeated freeze-thaw cycles. Samples can be stored at –20 °C if not analyzed immediately.
Assay Procedure Overview
- Prepare standards (dilute from 1080 to 60 pg/mL).
- Add 40 μL sample diluent + 10 μL sample to wells.
- Incubate for 30 minutes at 37 °C.
- Wash the plate 5 times with diluted wash buffer.
- Add 50 μL HRP-conjugated antibody to each well.
- Incubate again at 37 °C for 30 minutes.
- Wash again.
- Add 50 μL each of Chromogen A and B. Incubate 15 minutes at 37 °C in the dark.
- Add 50 μL stop solution; measure absorbance at 450 nm within 15 minutes.
Data Analysis
- A standard curve is constructed using log-log plot of OD vs. concentration.
- Sample concentrations are extrapolated from the curve and multiplied by dilution factor.
- All standards and samples should be run in duplicate.
