Protocol Overview:

  1. Prepare TrypLE™ Solution:
    • Dilute TrypLE™ Select 10X to 1X with a balanced salt solution containing 1 mM EDTA.
  2. Wash Cells:
    • Wash adherent cells with PBS or equivalent to remove serum that may inhibit enzyme activity.
  3. Apply TrypLE™:
    • Add the diluted TrypLE™ solution to the cell culture to cover the monolayer.
  4. Incubate:
    • Incubate at 37°C for 2–5 minutes, monitoring for cell detachment.
  5. Neutralize:
    • Dilute TrypLE™ Select with complete growth medium to inactivate the enzyme.
  6. Centrifuge and Resuspend:
    • Centrifuge cells and resuspend in fresh medium for further use.
