Assay Principle
In this colorimetric assay, LDH catalyzes the oxidation of lactic acid to pyruvate. Pyruvate reacts with 2,4-dinitrophenylhydrazine to form pyruvate dinitrophenylhydrazone, a red-brown complex in alkaline solution. The intensity of the color, measured with a microplate reader, is directly proportional to LDH activity in the sample.
