Assay Principle
The Abbexa HPR ELISA Kit is based on sandwich enzyme-linked immunosorbent assay (ELISA) technology:
1️⃣ Capture Antibody Pre-Coating – The microplate is pre-coated with an HPR-specific antibody.
2️⃣ Sample & Standard Addition – Standards, diluted samples, and control solutions are added to designated wells and incubated.
3️⃣ Binding & Detection – Biotin-conjugated secondary antibody is added, followed by HRP-conjugated reagent to amplify the signal.
4️⃣ Enzymatic Reaction – TMB substrate is added, producing a blue color proportional to the amount of HPR present.
5️⃣ Colorimetric Readout at 450 nm – Stop solution is added, turning the solution yellow. The intensity is measured using a microplate reader.

Assay Procedure
1️⃣ Equilibrate all reagents and samples to room temperature (18-25°C) before starting the assay.
2️⃣ Set up standard, test samples, and control wells on the pre-coated plate.
3️⃣ Add 100 µL of each diluted sample/standard into their respective wells. Incubate for 1 hour at 37°C.
4️⃣ Add 100 µL of Detection Reagent A to each well and incubate for 1 hour at 37°C.
5️⃣ Wash plate 3 times, then add 100 µL of Detection Reagent B. Incubate for 30 minutes at 37°C.
6️⃣ Wash plate 5 times, then add 90 µL of TMB Substrate and incubate for 10-20 minutes at 37°C.
7️⃣ Stop the reaction with 50 µL of Stop Solution and measure optical density (OD) at 450 nm.
Results Calculation & Interpretation
- Average the O.D.450 duplicate readings for each sample and standard.
- Subtract the blank (zero) O.D.450 reading from each value.
- Construct a standard curve by plotting O.D.450 vs. concentration (pg/mL).
- Interpolate sample HPR concentrations from the standard curve.
