    Y-27632 is a highly potent, cell-permeable, and selective Rho-associated coiled-coil-containing protein kinase (ROCK) inhibitor. It competitively binds to the ATP-binding site of ROCK-I (p160ROCK) and ROCK-II, key effectors in the RhoA/ROCK signaling pathway. Supplied as a convenient InSolution™ preparation, this reagent is suitable for direct use in cell culture and biochemical assays without reconstitution.

    Biochemical Activity & Selectivity

    • Primary Target: ROCK-I (p160ROCK) and ROCK-II
    • Ki: 140 nM for ROCK-I
    • IC₅₀: 700 nM for phenylephrine-induced Ca²⁺ sensitization in aortic smooth muscle
    • Selectivity Profile:
      • 10–50× lower affinity for PKCε than for ROCK
      • No activity against p21-activated kinase (PAK) up to 100 µM

    Mechanism: ATP-competitive inhibition of ROCK catalytic domains, resulting in downstream inhibition of myosin light chain phosphorylation, stress fiber formation, and cellular contractility.
