Y-27632 is a highly potent, cell-permeable, and selective Rho-associated coiled-coil-containing protein kinase (ROCK) inhibitor. It competitively binds to the ATP-binding site of ROCK-I (p160ROCK) and ROCK-II, key effectors in the RhoA/ROCK signaling pathway. Supplied as a convenient InSolution™ preparation, this reagent is suitable for direct use in cell culture and biochemical assays without reconstitution.

Biochemical Activity & Selectivity
- Primary Target: ROCK-I (p160ROCK) and ROCK-II
- Ki: 140 nM for ROCK-I
- IC₅₀: 700 nM for phenylephrine-induced Ca²⁺ sensitization in aortic smooth muscle
-
Selectivity Profile:
- 10–50× lower affinity for PKCε than for ROCK
- No activity against p21-activated kinase (PAK) up to 100 µM
Mechanism: ATP-competitive inhibition of ROCK catalytic domains, resulting in downstream inhibition of myosin light chain phosphorylation, stress fiber formation, and cellular contractility.
Applications in Research
- Cell migration and cytoskeletal remodeling
- Smooth muscle contraction inhibition
- Apoptosis and survival pathway studies
- Stem cell culture and reprogramming (e.g., maintenance of pluripotent stem cells)
- Wound healing and endothelial barrier function
- Cancer metastasis and invasion models
Advantages of InSolution™ Format
- Time-saving – no reconstitution needed
- Stability – aliquot and freeze for consistent dosing
- Reliability – consistent concentration, tested by HPLC, NMR, and MS
- Convenience – delivered in aqueous format, immediately compatible with culture systems
Storage & Handling
- Store at −20°C
- Protect from light to maintain compound stability
- After thawing, aliquot and refreeze to avoid multiple freeze-thaw cycles
- Product is stable for at least 6 months under recommended conditions
