Protocol Overview:

  1. Pre-Warm TrypLE™:
    • Warm TrypLE™ Express to room temperature before use.
  2. Wash Cells:
    • Wash adherent cells with PBS or equivalent to remove serum, which can inhibit enzyme activity.
  3. Add TrypLE™:
    • Add enough TrypLE™ Express to cover the cell monolayer.
  4. Incubate:
    • Incubate at 37°C, monitoring under a microscope until cells detach (2–5 minutes).
  5. Neutralize:
    • Dilute TrypLE™ Express with complete growth medium (no trypsin inhibitor required).
  6. Centrifuge and Resuspend:
    • Centrifuge cells, discard the supernatant, and resuspend in fresh medium.
