Protocol Overview:
- Pre-Warm TrypLE™:
- Warm TrypLE™ Express to room temperature before use.
- Wash Cells:
- Wash adherent cells with PBS or equivalent to remove serum, which can inhibit enzyme activity.
- Add TrypLE™:
- Add enough TrypLE™ Express to cover the cell monolayer.
- Incubate:
- Incubate at 37°C, monitoring under a microscope until cells detach (2–5 minutes).
- Neutralize:
- Dilute TrypLE™ Express with complete growth medium (no trypsin inhibitor required).
- Centrifuge and Resuspend:
- Centrifuge cells, discard the supernatant, and resuspend in fresh medium.
