Mode of Action & Resistance
Mechanism of Action:
- Penicillin G inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs).
- This prevents cross-linking of peptidoglycan chains, leading to cell lysis and bacterial death.
- Most effective against actively dividing bacterial cells.
Mode of Resistance:
- Bacterial β-lactamases hydrolyze Penicillin G, inactivating its function.
- Some bacteria alter PBPs, reducing antibiotic binding and conferring resistance.
- Resistance is common in penicillinase-producing Staphylococcus spp. and Enterococcus spp..
Antimicrobial Spectrum:
✔ Gram-Positive Bacteria (Streptococcus spp., Staphylococcus spp., Bacillus spp.)
✔ Gram-Negative Bacteria (Neisseria meningitidis, Haemophilus influenzae)
✔ Anaerobic Bacteria (Clostridium spp., Actinomyces spp.)
Applications
🔬 Microbiology & Bacterial Culture – Prevents contamination in bacterial culture media & microbiological assays
🦠 Mammalian Cell Culture Studies – Used in cell culture media to prevent bacterial contamination
⚗️ Antibiotic Resistance Research – Evaluates Penicillin G susceptibility in bacterial strains
🧪 Pharmaceutical & Drug Development – Used in antibiotic formulation & bacterial inhibition studies
🏭 Food & Beverage Safety Testing – Screens for penicillin-resistant bacterial contamination
Preparation & Storage Guidelines
1️⃣ Preparation of Penicillin G Stock Solution
- Dissolve 100 mg/mL in sterile water or PBS.
- Filter sterilize before use (do not autoclave).
- Store at 2–8°C (short-term) or -20°C (long-term) for extended stability.
2️⃣ Recommended Working Concentrations
- Bacterial Selection & Growth Inhibition: 10–100 µg/mL
- Mammalian Cell Culture Protection: 50–100 U/mL
3️⃣ Storage & Handling
- Keep tightly sealed in a dry, ventilated environment.
- Protect from moisture & high temperatures to prevent degradation.
- Avoid repeated freeze-thaw cycles for optimal stability.
