Preparation Instructions

1. Suspend 42g of powder in 1 liter of distilled or deionized water.
2. Heat to boiling, stirring until completely dissolved.
3. Autoclave at 121°C for 15 minutes.
4. Cool the medium to 45-50°C.
5. Add rehydrated selective supplements as needed:
   • T.S.C. Supplement (D-Cycloserine).
   • Kanamycin/Polymyxin B Supplement.
   • D-Cycloserine 4-MUP Supplement for fluorescence detection.
6. Mix thoroughly and pour into sterile Petri dishes.

Interpretation of Results

• Clostridium perfringens: Black colonies due to H₂S production.
• On TSC Agar with MUP: Light blue fluorescence under UV indicates Clostridium perfringens.
• Other Organisms:
  • Escherichia coli: Inhibited.
  • Bacillus subtilis: Inhibited.
