This medium, when supplemented with Polymyxin B and Egg Yolk Emulsion, ensures the detection of low numbers of B. cereus in the presence of large amounts of background flora, adhering to standards such as ISO 21871 and FDA-BAM.

Preparation Instructions

1. Suspend 41g of powder in 1 liter of distilled or deionized water.
2. Heat to boil, shaking to ensure complete dissolution.
3. Autoclave at 121°C for 15 minutes.
4. Cool to 45-50°C and aseptically add:
   • Rehydrated contents of 2 vials of Polymyxin B Supplement (Ref. 81016).
   • 50 mL of Egg Yolk Emulsion (Ref. 80124 or 80219).
5. Mix thoroughly and pour into sterile Petri dishes.

Colony Characteristics

• Bacillus cereus:
  • Blue colonies due to bromothymol blue.
  • Lecithinase-positive, indicated by a precipitation halo around colonies.
• Other Bacteria:
  • Mannitol fermenters form yellow colonies.
  • Non-lecithinase producers lack precipitation halos.
