High-Potency Glycopeptide Antibiotic for Stable Selection in Mammalian, Insect, Yeast, Bacterial, and Plant Cells
Gibco™ Zeocin™ Selection Reagent is a sterile-filtered liquid formulation of phleomycin D1, a glycopeptide antibiotic produced by Streptomyces verticillus. Zeocin™ belongs to the bleomycin/phleomycin family and exerts its cytotoxic activity by binding to and cleaving DNA, which leads to rapid cell death in non-resistant cells. This mechanism makes it highly effective for selecting genetically modified cells that express a Zeocin resistance gene (Sh ble), commonly included in many expression and transfection vectors.
Supplied as a 100 mg/mL solution in sterile water, Zeocin™ is suitable for use in a wide range of cell types and organisms, including mammalian cells, insect cells, yeast, bacteria, and plant cultures.
Key Features and Benefits
-
High-Activity Glycopeptide Antibiotic
Contains phleomycin D1, which causes double-stranded DNA breaks, leading to rapid cytotoxicity in sensitive cells. -
Broad Compatibility
Effective for selection in mammalian, insect, yeast, bacterial, and plant systems—ideal for multi-platform workflows. -
Rapid and Reliable Selection
Enables efficient elimination of non-transfected or non-transformed cells, ensuring only stably modified cells survive. -
Convenient Ready-to-Use Format
Supplied as a 100 mg/mL sterile-filtered solution, eliminating the need for reconstitution or filtration. -
Stable and Easy to Store
Ships on ice and should be stored at –5°C to –20°C, with a long shelf life under proper conditions.
Applications
- Selection of mammalian and insect cells carrying the Sh ble resistance gene
- Generation of stable cell lines following plasmid transfection
- Antibiotic selection in yeast and E. coli transformation workflows
- Transgenic plant selection using Zeocin-resistant vectors
- Dual-antibiotic selection with other agents like Hygromycin B or Blasticidin
Recommended Working Concentrations
Organism / System | Working Concentration (µg/mL) |
---|---|
Mammalian cells | 50–400 µg/mL |
Insect cells | 50–200 µg/mL |
Yeast cells | 75–100 µg/mL |
Bacterial cells | 25–50 µg/mL |
Plant cells | 50–100 µg/mL |
A kill curve is recommended to determine the optimal concentration for your specific cell type.
