Thermo Scientific™ Bond-Breaker™ TCEP Solution, Neutral pH is a ready-to-use, stable 0.5 M solution of tris(2-carboxyethyl)phosphine, commonly known as TCEP. Usage Considerations
Reduction can occur efficiently over a broad pH and temperature range. Most proteins can be reduced without a denaturant, although a denaturant such as guanidine hydrochloride may help expose internal disulfide bonds. Urea is not recommended because it can form cyanates that react with sulfhydryl groups.
Metal contact should be avoided because metals can reduce TCEP activity. Including 5–20 mM EDTA in the sample buffer can help prevent reoxidation of reduced sulfhydryl groups by divalent metals such as zinc, copper, and magnesium. Reduced samples should ideally be used immediately because disulfide bonds may reform over time.
