Thermo Scientific™ Pierce™ Ni-NTA Magnetic Agarose Beads provide a fast and convenient magnetic affinity purification method for His-tagged recombinant proteins. The beads are designed for both manual benchtop purification and automated high-throughput workflows using magnetic particle processors such as the Thermo Scientific™ KingFisher™ Flex system.
The beads are incubated with clarified cell lysate containing the His-tagged target protein, magnetically separated from the supernatant, washed to remove nonspecifically bound proteins, and then eluted using an appropriate elution buffer. This workflow supports rapid purification, screening of expression conditions, and small-scale recombinant protein purification.

Technology Overview
The beads consist of highly crosslinked agarose embedded with magnetite particles and functionalized with a covalently attached nitrilotriacetic acid, NTA, chelator charged with divalent nickel ions. The nickel-charged NTA ligand interacts with histidine residues in His-tagged proteins, allowing selective capture and purification from cell lysates.
