The Human Suppressors of Cytokine Signaling 1 (SOCS1) ELISA Kit is a highly sensitive immunoassay designed for the quantitative measurement of Human SOCS1 antigen in biological samples including serum, plasma, cell culture supernatants, tissue lysates, and other biological fluids.
SOCS1 is a key intracellular protein involved in regulating cytokine signaling, particularly through the Janus kinase / signal transducer and activator of transcription (JAK/STAT) pathway. Assay Principle (Sandwich ELISA)
- Microplate wells are pre-coated with antibodies specific to Human SOCS1.
- SOCS1 antigen in the sample binds to the immobilized antibody.
- After washing, an HRP-conjugated antibody binds to the captured antigen.
- Addition of TMB chromogenic substrate produces a blue color.
- The reaction is stopped using an acid stop solution, turning the color yellow.
- Absorbance is measured at 450 nm, and SOCS1 concentration is calculated from a standard curve.

Sample Preparation
Serum
Collect blood in a plain tube and allow it to clot for 10–20 minutes at room temperature. Centrifuge at 2000–3000 rpm for 20 minutes and collect the serum.
Plasma
Collect blood using EDTA or citrate anticoagulant tubes, incubate for 10–20 minutes, centrifuge at 2000–3000 rpm for 20 minutes, and collect the plasma supernatant.
Urine and Other Fluids
Collect samples in sterile tubes and centrifuge at 2000–3000 rpm for 20 minutes to obtain the clear supernatant.
Cell Culture Samples
Centrifuge culture supernatants at 2000–3000 rpm for 20 minutes and collect the clarified supernatant.
Tissue Samples
Homogenize tissues in PBS (pH 7.4) under cold conditions and centrifuge at 2000–3000 rpm for 20 minutes. Collect the supernatant for analysis.
