Liofilchem® Brucella Supplement is a freeze-dried mixture of selective antibiotics and antifungals formulated to enhance the isolation of Brucella species by suppressing the growth of contaminating microorganisms.
When added to Brucella Agar Base (ref. 610079 / 620079), Columbia Agar Base (ref. 610013 / 620013), or Blood Agar Base N.2 (ref. 610188 / 620188), the supplement provides the selectivity necessary for recovery of Brucella spp. from milk, food, or veterinary samples while maintaining high recovery rates for slow-growing target organisms.
Key Features
- Selective supplement designed specifically for the isolation of Brucella spp.
- Broad inhibitory range—suppresses Gram-positive, Gram-negative, and fungal contaminants.
- Freeze-dried, ready-to-reconstitute format for convenient use and reproducibility.
- Each vial sufficient for 500 mL of medium.
- Enhances Brucella recovery when used with 5–10 % inactivated horse serum and 1–5 % sterile glucose.
- Validated and quality-controlled under ISO-certified manufacturing standards.
Composition per Vial
| Component | Per Bottle | Per Litre of Medium | Function |
|---|---|---|---|
| Polymyxin B | 2 500 IU | 5 000 IU | Inhibits Gram-negative contaminants |
| Bacitracin | 12 500 IU | 25 000 IU | Inhibits Gram-positive bacteria |
| Cycloheximide | 50 mg | 100 mg | Antifungal agent |
| Nalidixic Acid | 2.5 mg | 5 mg | Quinolone active against Gram-negatives |
| Nystatin | 50 000 IU | 100 000 IU | Antifungal activity |
| Vancomycin | 10 mg | 20 mg | Inhibits cell-wall synthesis in Gram-positives |
Principle of the Method
The slow growth of Brucella species requires selective agents to minimize interference from faster-growing flora.
- Polymyxin B and Nalidixic acid act on Gram-negative bacteria.
- Bacitracin and Vancomycin inhibit Gram-positive organisms.
-
Cycloheximide and Nystatin suppress yeasts and molds.
This balanced antibiotic combination creates a selective environment ideal for Brucella isolation and subsequent identification.
Preparation and Use
- Reconstitution: Aseptically add 10 mL of a 1 : 1 mixture of methanol and sterile distilled water to one vial.
- Dissolve completely by gentle shaking and incubate at 36 ± 1 °C for 10–15 min.
- Add entire contents (10 mL) to 500 mL of Brucella Agar Base or other recommended basal medium cooled to 45–50 °C, already supplemented with 5–10 % inactivated horse serum and 1–5 % sterile glucose solution.
- Mix gently and pour into Petri dishes.
Quality Control
| Test Organism | ATCC No. | Expected Result |
|---|---|---|
| Brucella abortus | 4315 | Good growth |
| Staphylococcus aureus | 25923 | Inhibited |
| Escherichia coli | 25922 | Inhibited |
