The Invitrogen™ p53 Monoclonal Antibody (Clone DO-7) is a highly specific, mouse-derived IgG2b antibody designed to detect wild-type p53 protein in various research applications, including Western blotting (WB), immunocytochemistry (ICC/IF), immunohistochemistry (IHC), flow cytometry (Flow), immunoprecipitation (IP), and chromatin immunoprecipitation (ChIP). This antibody is unconjugated and is supplied in liquid form at a concentration of 0.05 mg/mL in PBS with 0.05% sodium azide as a preservative.

Key Features:

  • Clone: DO-7
  • Host/Isotype: Mouse / IgG2b, κ
  • Target: Human p53 (Cellular tumor antigen p53)
  • Reactivity: Human, Bovine (published reactivity with Human, Mouse, Rat)
  • Applications: WB, ICC/IF, IHC (P), IP, Flow Cytometry, ChIP
  • Conjugate: None (Unconjugated)
  • Purification: Protein A affinity chromatography
  • Storage: –20°C (long-term); shipped on wet ice
  • Volume: 500 µL
  • Catalog Number: MA5-12557
  • RRID: AB_10989883

Applications & Performance:

  • Western Blot (WB): 1:500 – 1:4,000 dilution
  • Immunocytochemistry (ICC/IF): 1:20 – 1:200 dilution
  • Immunoprecipitation (IP): 1:50 – 1:100 dilution
  • Immunohistochemistry (Paraffin): 1:100 – 1:200 dilution
  • Flow Cytometry: 1:50
  • ChIP Assay: 1 µg/mL

Product Validation:

This antibody has been verified through advanced verification methods, including knockout validation and extensive literature support. It has been referenced in over 170 peer-reviewed publications and used in high-content screening platforms, confirming its reliability across multiple assay systems.

Biological Background:

The p53 protein is a crucial tumor suppressor and transcription factor that responds to DNA damage by inducing cell cycle arrest or apoptosis. It is composed of distinct functional domains, including a transactivation domain, DNA-binding domain, and oligomerization domain. Mutations in the TP53 gene are implicated in more than 50% of human cancers. The DO-7 clone recognizes both wild-type and mutant forms of human p53 and is widely used in cancer diagnostics and basic research.

Storage & Handling:

  • Store at –20°C. Avoid repeated freeze-thaw cycles.
  • For optimal performance, briefly centrifuge the antibody vial before use.

Recommended Use:

Ideal for researchers studying cell cycle regulation, apoptosis, tumor suppressor pathways, DNA damage response, and oncogenesis. Suitable for use in cancer research, drug screening, and biomarker discovery involving p53 expression and mutation status.

Application: Western Blot, immunocytochemistry, Flow Cytometry, Immunohistochemistry, Immunoprecipitation (IP)
Storage Temperature: 2-8°C
