Principle of the Assay

The Vircell Parainfluenza ELISA Kit employs an indirect ELISA methodology:

  1. Antigen Binding:
    • Parainfluenza antigens are adsorbed onto the polystyrene wells of the ELISA plate.
  2. Sample Interaction:
    • Antibodies in the sample bind to the adsorbed antigens. Unbound immunoglobulins are washed off.
  3. Conjugate Reaction:
    • An enzyme-labeled anti-human globulin binds to the antigen-antibody complex.
  4. Signal Development:
    • TMB substrate is added, producing a blue-colored solution. Upon stopping the reaction, the color changes to yellow.
  5. Quantification:
    • The optical density (OD) is measured at 450 nm, and antibody levels are determined based on OD values.

Applications and Use Cases

1. Clinical Diagnostics:

  • Acute Infection Detection:
    • Detect IgM antibodies to confirm recent Parainfluenza virus infections.
  • Immunity Assessment:
    • Measure IgG antibodies to evaluate immunity from past infections or vaccination.

2. Epidemiological Studies:

  • Seroprevalence Monitoring:
    • Assess Parainfluenza virus exposure in populations.
  • Outbreak Investigations:
    • Identify infection patterns during outbreaks of respiratory diseases.

3. Vaccine Research:

  • Post-Vaccination Studies:
    • Evaluate the immune response and efficacy of Parainfluenza vaccines.
  • Efficacy Testing:
    • Measure antibody production after vaccination.

4. Laboratory Research:

  • Immunological Studies:
    • Explore immune responses to Parainfluenza virus infections.
  • Antibody Profiling:
    • Differentiate IgG and IgM responses to Parainfluenza types 1, 2, and 3.
