Thermo Scientific™ Maxima H Minus Reverse Transcriptase (RT) is a next-generation, thermostable reverse transcriptase engineered through in vitro evolution of Moloney Murine Leukemia Virus (M-MuLV) RT. The enzyme maintains both RNA-dependent and DNA-dependent polymerase activities while lacking RNase H activity due to a targeted mutation in the RNase H domain. This unique design enables efficient synthesis of high-quality, full-length cDNA up to 20 kb, even from complex or highly structured RNA templates.

With up to 50-fold higher processivity than wild-type M-MuLV RT, Maxima H Minus RT delivers faster synthesis rates, exceptional robustness, and consistent performance across a wide range of RNA inputs. Its enhanced thermostability allows reactions to be performed at temperatures up to 65 °C, improving specificity and reducing the impact of RNA secondary structures.

Key Features

  • High thermostability – retains ~90% activity after 60 minutes at 50 °C
  • Active up to 65 °C – ideal for GC-rich or highly structured RNA templates
  • RNase H–minus formulation – enables synthesis of long, intact cDNA up to 20 kb
  • Ultra-high processivity – approximately 50× higher than wild-type M-MuLV RT
  • Rapid reaction kinetics – complete cDNA synthesis in 15–30 minutes
  • Exceptional sensitivity – reproducible results from 1 pg to 5 µg total RNA
  • Enhanced inhibitor tolerance – resistant to guanidine, formamide, and ethanol
  • Modified nucleotide compatibility – suitable for labeled cDNA applications

Applications

  • First-strand cDNA synthesis for RT-PCR and RT-qPCR
  • Reverse transcription at elevated temperatures to overcome RNA secondary structures
  • cDNA synthesis for cloning and gene expression analysis
  • Preparation of labeled cDNA probes for microarray applications
  • RNA analysis using primer extension methods
