The PolyCAT A™ Column is a high-performance weak cation-exchange (WCX) chromatography column specifically designed for the separation of proteins, histones, monoclonal antibodies, and hemoglobin variants. It is built by covalently attaching poly(aspartic acid) to silica, providing sharp peak resolution, high recovery, and minimal peak tailing.

Available in various dimensions with 2 µm particle sizes and multiple pore size options (-03, -10, -15), PolyCAT A™ offers superior selectivity and efficiency for proteins with an isoelectric point (pI) above 6.0. It is commonly used in biotechnology, clinical chemistry, and pharmaceutical applications for high-resolution protein analysis and characterization.

Key Features & Benefits

1. High-Resolution Cation-Exchange Separation

  • Poly(aspartic acid)-coated silica for efficient protein separation with sharp peaks.
  • Strong retention of positively charged proteins, ensuring high-resolution elution.
  • Minimal peak tailing, enhancing detection sensitivity and reproducibility.

2. Versatile Applications for Protein & Variant Analysis

  • Protein variant analysis – Ideal for deamidation, PEGylation, and desialylation studies.
  • Hemoglobin variant analysis in clinical chemistry labs.
  • Monoclonal antibody (mAb) characterization for biopharmaceutical research.
  • Histone separation and analysis in epigenetic research.

3. Multiple Pore Sizes & Configurations for Specific Applications

  • 2 µm particle size for ultra-high-resolution separations.
  • Available pore size options (-03, -10, -15) for optimal selectivity.
  • Recommended pore size ≥ 1000 Å for proteins >20 kDa.

4. Optimized for pH-Dependent Protein Elution

  • Operates best at pH 6-7 for optimal cation-exchange separation.
  • Gradient elution with unbuffered acetic acid permits elution in volatile solvents.
  • Avoid prolonged exposure to pH above 8 to maintain column longevity.

Column Use & Maintenance

Initial Use & Conditioning

  • Flush new columns with 15 column volumes of water before first use.
  • Condition with a 40 mM EDTA.2Na solution (filtered, unadjusted pH) at low flow rate (20-24 hours).
  • Alternatively, use a 0.2 M NaH₂PO₄ + 0.3 M sodium acetate solution for 2 hours.

Routine Use & Column Care

  • Proteins elute with salt and/or pH gradients (pH 6-7 recommended).
  • Filter mobile phases and samples to avoid clogging the column frit.
  • Avoid prolonged exposure to pH above 8 to prevent degradation of the stationary phase.
  • Store in 100% mobile phase A (short-term) or water (long-term at 4°C).

Column Regeneration & Longevity

  • Invert column and flush with 40 mM EDTA.2Na overnight after every 250 injections.
  • For heavy usage (500+ injections), replenish with bulk PolyCAT A™ material.
  • Use at room temperature (20-25°C) to preserve column integrity.
