Membrane-potential-sensitive dye for mitochondrial function analysis by flow cytometry
The Invitrogen™ MitoProbe™ DiOC₂(3) Assay Kit is a convenient, ready-to-use solution for assessing mitochondrial membrane potential (ΔΨm) in live eukaryotic cells using flow cytometry. The kit contains DiOC₂(3), a lipophilic, cationic cyanine dye that selectively accumulates in mitochondria with active membrane potentials. In polarized mitochondria, DiOC₂(3) aggregates, producing a bright red fluorescence signal; as membrane potential decreases, the dye disperses, causing a shift from red to green fluorescence.
The kit also includes CCCP (carbonyl cyanide 3-chlorophenylhydrazone), a mitochondrial membrane potential disruptor, for use as a control in experiments. This combination enables accurate detection of changes in mitochondrial polarization in response to treatments, apoptosis induction, or metabolic alterations.
