The Invitrogen™ IL-17A Monoclonal Antibody (eBio64DEC17), conjugated to APC (Allophycocyanin), is a highly specific antibody designed for detecting human IL-17A in intracellular staining followed by flow cytometric analysis. IL-17A is a cytokine secreted by Th17 cells and plays a critical role in inflammatory responses and immune regulation. This antibody is validated for intracellular staining and offers bright fluorescence for reliable detection.

Key Features:

  1. Target and Specificity:
    • Recognizes human IL-17A, a 32 kDa cytokine involved in promoting inflammation and immune responses.
    • Neutralizing antibody with high specificity for IL-17A.
  2. Clone and Conjugate:
    • Clone: eBio64DEC17, ensuring reliable and reproducible performance.
    • Conjugate: APC, providing bright and stable fluorescence with excitation at 651 nm and emission at 660 nm.
  3. Validated Applications:
    • Optimized for intracellular staining and flow cytometry to study IL-17A expression in polarized Th17 cells and other immune cells.
  4. Bright and Stable Fluorescence:
    • APC fluorochrome offers high signal intensity for clear separation of IL-17A-positive and negative populations.
  5. Ready-to-Use:
    • Pre-titrated for ease of use, with 5 µL (0.06 µg) sufficient for each test.

Applications:

  1. Intracellular Staining:
    • Detect IL-17A expression in CD4+ T cells, including Th17-polarized cells.
    • Analyze cytokine production in immune cells under stimulated conditions.
  2. Flow Cytometry:
    • Ideal for use in multicolor panels to identify IL-17A-producing cells in human samples.
  3. Functional Assays:
    • Investigate the role of IL-17A in inflammatory responses, autoimmunity, and disease models.
  4. Th17 Research:
    • Study IL-23-dependent Th17 differentiation pathways and their role in inflammatory diseases.
  5. Inflammation Studies:
    • Examine IL-17A involvement in conditions such as rheumatoid arthritis, asthma, multiple sclerosis, and psoriasis.

Key Benefits:

  1. High Specificity and Sensitivity:
    • Detects IL-17A with minimal cross-reactivity or background.
  2. Bright Fluorescence:
    • APC provides a strong and stable signal for clear identification of IL-17A-positive cells.
  3. Reproducible Results:
    • Pre-titrated antibody ensures consistency and reliability across experiments.
  4. Flexibility:
    • Compatible with various fixation and permeabilization protocols for intracellular staining.
  5. Validated for Research:
    • Optimized for studying IL-17A production in human immune cells using flow cytometry.

Usage Recommendations:

  1. Sample Preparation:
    • Polarize human peripheral blood CD4+ T cells under Th17 conditions.
    • Stimulate cells using appropriate reagents (e.g., Cell Stimulation Cocktail).
  2. Staining Protocol:
    • Use the Intracellular Fixation & Permeabilization Buffer Set for effective staining.
    • Add 5 µL (0.06 µg) of antibody per 100 µL of cell suspension.
  3. Flow Cytometry:
    • Analyze samples using a flow cytometer equipped with a red laser (633–647 nm).
  4. Fixation:
    • Fix samples in IC Fixation Buffer to preserve fluorescence for up to 3 days.

Storage and Handling:

  • Store at 4°C in the dark to protect the APC fluorochrome.
  • Do not freeze, as freezing may degrade the antibody or fluorochrome.

Target Information:

  • IL-17A:
    • A pro-inflammatory cytokine secreted by Th17 cells.
    • Plays a key role in autoimmune diseases, inflammation, and immune responses against fungal and bacterial infections.
    • Stimulates fibroblasts, epithelial cells, and endothelial cells to produce cytokines such as IL-6, IL-8, and G-CSF.
