Assay Principle

This kit uses a double-antibody sandwich method. The microplate is pre-coated with a monoclonal antibody specific to Human HbC. After sample addition, HRP-conjugated anti-HbC antibodies bind to the captured antigen. A TMB substrate reacts with HRP to produce a blue color, which turns yellow upon addition of the stop solution. The absorbance is read at 450 nm, and the concentration is determined using a standard curve.

Assay Procedure Overview

  1. Prepare standard dilutions (180 → 10 ng/mL).
  2. Add 40 μL sample diluent + 10 μL sample to each well.
  3. Incubate 30 minutes at 37 °C, sealed.
  4. Wash 5× using diluted wash buffer.
  5. Add 50 μL HRP-conjugate, incubate again.
  6. Wash and add 50 μL each of Chromogen A & B. Incubate in dark for 15 minutes.
  7. Add 50 μL stop solution. Measure absorbance at 450 nm within 15 minutes.
