The Thermo Scientific™ S1 Nuclease is a single-strand–specific endonuclease that selectively degrades single-stranded nucleic acids (DNA and RNA), releasing 5′-phosphoryl mono- and oligonucleotides. With approximately five times greater activity on DNA than RNA, it is a powerful tool for nucleic acid research and genome editing workflows.
In addition to single-stranded substrates, S1 Nuclease cleaves double-stranded DNA (dsDNA) at single-stranded regions, such as nicks, gaps, mismatches, or loops, and exhibits 3′-phosphomonoesterase activity. The enzyme is a glycoprotein containing 18% carbohydrate, contributing to its structural stability and functionality.
