The Invitrogen™ Glutathione Peroxidase (GSH-Px) Activity Kit provides a highly sensitive and reproducible method for quantifying GSH-Px activity in cell culture supernatants, plasma, serum, tissue homogenates, and whole cells. GSH-Px is a critical antioxidant enzyme that protects cells from oxidative damage by catalyzing the reduction of hydrogen peroxide and organic hydroperoxides, using reduced glutathione (GSH) as a cofactor.
In this assay, GSH-Px activity is determined by measuring the consumption of GSH. Reduced glutathione reacts with dinitrobenzoic acid (DTNB) to produce 5-thio-dinitrobenzoic acid anion, a stable yellow product. The absorbance of this product is measured at 412 nm to calculate GSH levels, which correlate with enzyme activity.

Assay Principle
- GSH-Px in the sample catalyzes the reduction of hydrogen peroxide or organic hydroperoxides using GSH as a cofactor.
- The residual GSH reacts with DTNB, producing a yellow-colored 5-thio-dinitrobenzoic acid anion.
- The absorbance at 412 nm is proportional to the amount of residual GSH, which inversely reflects GSH-Px activity.
