    The Invitrogen™ EnzChek™ Cellulase Substrate is a simple, rapid, and highly sensitive assay substrate for the quantitative measurement of cellulase activity. Designed for both routine and high-throughput applications, this substrate enables accurate results in 30 minutes or less with minimal steps.

    Unlike complex, multistep cellulase assays, the EnzChek™ substrate requires only enzyme addition, incubation, and fluorescence or absorbance reading. This makes it ideal for laboratories looking to increase throughput without compromising sensitivity.

    Principle of the Assay

    The assay uses a blue-fluorescent substrate with excitation/emission maxima at ~339/452 nm. In the presence of cellulase, the substrate is hydrolyzed, releasing highly fluorescent fragments proportional to enzyme activity.

    While optimized for fluorescence detection (providing maximum sensitivity, as low as 40 μU/mL cellulase), the substrate can also be used in a colorimetric assay with slightly reduced sensitivity.
