The Liofilchem® Coagulase Test is a high-quality diagnostic kit designed to detect the coagulase enzyme produced by Staphylococcus aureus. This test uses lyophilic rabbit plasma with EDTA to accurately and efficiently identify coagulase-positive Staphylococcus aureus, a key diagnostic marker for pathogenic strains. The kit is specifically formulated for in vitro diagnostic use in clinical, microbiological, and research laboratories.

    Key Features

    1. Comprehensive Kit Components:
      • Includes 5 vials, each containing 4 mL of lyophilic rabbit plasma, allowing for multiple tests per batch.
      • Includes detailed instruction sheet for standardized procedures.
    2. High Sensitivity and Specificity:
      • Reliably detects coagulase-positive Staphylococcus aureus, a critical diagnostic marker in clinical microbiology.
      • Tested with quality control organisms, ensuring accuracy:
        • Positive Control: Staphylococcus aureus ATCC 25923
        • Negative Control: Escherichia coli ATCC 25922
    3. Validated Testing Method:
      • Based on the conversion of fibrinogen to fibrin, mediated by coagulase produced by S. aureus.
      • EDTA chelates calcium, ensuring the reaction is specific to coagulase activity.
    4. Ready-to-Use Format:
      • Lyophilized rabbit plasma ensures ease of storage and reconstitution, providing consistent results.
    5. Flexible Testing Workflow:
      • Suitable for clinical laboratories, research applications, and quality control processes in microbiology.

    Kit Contents

    • 5 Vials of Rabbit Plasma (4 mL per vial)
    • Instruction Sheet

    Additional Items Required (Not Included)

    • Physiological Solution (Ref. 20095): For reconstitution of rabbit plasma.
    • Brain Heart Infusion Broth: For culture preparation before testing.

    Principle of the Test

The coagulase enzyme produced by Staphylococcus aureus reacts with fibrinogen in rabbit plasma, converting it into fibrin, resulting in clot formation. The EDTA in the formulation chelates calcium, ensuring the reaction is specific to coagulase activity without interference from other enzymatic processes.

    Step-by-Step Instructions

    1. Reconstitution:
      • Reconstitute one vial of COAGULASE TEST with 4 mL of physiological solution under aseptic conditions.
    2. Culture Preparation:
      • Pick colonies from selective media for S. aureus isolation.
      • Inoculate into Brain Heart Infusion Broth and incubate at 36 ± 1°C for 4-6 hours.
    3. Testing:
      • In a sterile tube, mix 0.5 mL of COAGULASE TEST reagent with 0.5 mL of culture broth.
      • Incubate the mixture at 36 ± 1°C.
    4. Result Interpretation:
      • Check for clot formation at intervals of 1, 2, 4, 8, and 24 hours.
      • Avoid incubating beyond 24 hours to prevent fibrinolysis, which may degrade the clot.
    5. Positive Result: Visible clot formation indicates the presence of coagulase-positive Staphylococcus aureus.
    6. Negative Result: No clot formation suggests the absence of coagulase activity.

    Applications and Use Cases

    1. Clinical Microbiology Diagnostics

    • Purpose:
      • Rapidly identify Staphylococcus aureus, a common pathogen causing skin infections, bloodstream infections, and pneumonia.
    • Use Case:
      • Confirm coagulase-positive S. aureus isolates from clinical samples such as blood, wounds, or respiratory secretions.

    2. Food and Beverage Testing

    • Purpose:
      • Screen for coagulase-positive Staphylococcus aureus, a major indicator of food contamination.
    • Use Case:
      • Quality assurance in food production facilities to prevent outbreaks caused by S. aureus toxin contamination.

    3. Pharmaceutical and Cosmetic Testing

    • Purpose:
      • Ensure product safety by identifying potential contamination with pathogenic S. aureus.
    • Use Case:
      • Routine microbiological testing of raw materials and finished products.

    4. Academic and Research Laboratories

    • Purpose:
      • Study coagulase activity in pathogenicity and virulence research.
    • Use Case:
      • Investigate mechanisms of S. aureus infection or develop new antibiotics targeting coagulase-producing strains.

    Advantages

    1. High Reliability:
      • Accurate differentiation of coagulase-positive S. aureus from other species.
    2. Ease of Use:
      • Ready-to-use, lyophilized rabbit plasma simplifies workflow and storage.
    3. Validated Performance:
      • Consistently high-quality results through stringent quality control with reference strains.
    4. Cost-Effective:
      • Multiple tests per vial reduce overall testing costs.
    5. Regulatory Compliance:
      • Conforms to global diagnostic standards for in vitro testing.

    Precautions

    • For in vitro diagnostic use only.
    • Use by properly trained personnel in approved laboratory settings.
    • Store at 2–8°C and avoid excessive temperature fluctuations.
    • Dispose of all used materials as per laboratory protocols for biohazard waste.

    Storage and Stability

    • Storage Conditions: 2–8°C in original packaging.
    • Shelf Life: Valid until the expiration date indicated on the label.
    • Signs of Deterioration: Do not use if any vial shows signs of contamination or degradation.

    Quality Control

Each batch is tested using the following strains:

    • Positive Control: Staphylococcus aureus ATCC 25923 (Clot Formation)
    • Negative Control: Escherichia coli ATCC 25922 (No Clot Formation)
