Reagents for Indole and Oxidase Tests for use with the A.F. Genital System (ref. 74156). This reagent kit is designed to support the proper execution of biochemical tests, microbial identification, and susceptibility testing of pathogenic urogenital microorganisms.
Description
The A.F. Genital System Reagent includes specialized reagents and tools to perform:
- Indole Test (Kovac’s Reagent) for biochemical characterization.
- Oxidase Test (Oxidase Test Stick) for the confirmation of Neisseria gonorrhoeae.
- Anaerobic Maintenance (Vaseline Oil) to ensure proper environmental conditions for specific wells.
These reagents are critical for obtaining accurate and reliable results from the A.F. Genital System panel.
Kit Contents
Component | Description | Quantity |
---|---|---|
Kovac's Reagent | 5% (w/v) p-dimethylaminobenzaldehyde in 25% hydrochloric acid and 75% isobutyl alcohol, for indole test. | 1 x 10 mL vial |
Oxidase Test Stick | Paper strips impregnated with 1% tetramethyl-p-phenylenadiamine dihydrochloride. | 2 x 50 strips |
Vaseline Oil | Pure sterile oil for anaerobic maintenance. | 2 x 50 mL bottles |
Empty Dropper Bottle | For optional use with reagents. | 1 bottle |
Method Principle
- Kovac’s Reagent:
- Used for the Indole Test in well 17-PRO, identifying microorganisms capable of tryptophan degradation to indole.
- Positive Result: Development of a red/pink layer.
- Oxidase Test Stick:
- Confirms the presence of Neisseria gonorrhoeae using liquid from well 22-NES.
- Positive Result: Development of a deep purple/blue color on the oxidase test strip.
- Vaseline Oil:
- Ensures anaerobic conditions for specific wells, excluding wells such as 6-TR/YE, 16-ESC, and 22-NES.
Applications
- Clinical Diagnostics:
- Used to identify and test the susceptibility of pathogens causing urogenital infections, including Neisseria gonorrhoeae and Trichomonas vaginalis.
- Industrial Microbiology:
- Suitable for quality control and contamination testing in clinical-grade materials.
- Research:
- Useful in studies involving urogenital microbiota and pathogen behavior.
Test Procedure
- Refer to the A.F. Genital System package insert for detailed instructions.
- Add Kovac's Reagent to well 17-PRO for the Indole Test.
- Use Oxidase Test Stick with liquid from well 22-NES to confirm N. gonorrhoeae.
- Seal wells with Vaseline Oil as specified for anaerobic maintenance.
Storage and Stability
- Temperature: Store at 2–8°C, away from light in its original packaging.
- Stability: Discard reagents if signs of contamination or deterioration are evident.
Precautions
- For in vitro diagnostic use only.
- Classified as hazardous; consult the Safety Data Sheet (SDS) for handling instructions.
- Handle using appropriate aseptic techniques and safety protocols.
Quality Control
Validate reagents using appropriate positive and negative control organisms:
- Positive Controls: Escherichia coli (Indole test), Neisseria gonorrhoeae (Oxidase test).
- Negative Controls: Organisms lacking indole production or oxidase activity.
Disposal
Dispose of used reagents and materials in accordance with local regulations for hazardous and biohazardous waste.
