Principle of the Method
- Voges-Proskauer Test:
- Detects the production of acetoin from glucose fermentation.
- Reagents Used: Alpha Naphtol and NaOH 40%.
- Positive Result: Pink to red color in well 10-VP.
- Indole Test:
- Identifies the ability of bacteria to produce indole from tryptophan degradation.
- Reagent Used: Kovac's Reagent.
- Positive Result: Formation of a red/pink layer in well 11-IND.
- Anaerobic Maintenance:
- Vaseline Oil ensures an oxygen-restricted environment for specific biochemical reactions (e.g., lysine, ornithine, arginine decarboxylation).
Reagents
Reagent | Composition |
---|---|
Alpha Naphtol | 6% (w/v) alpha naphtol dissolved in ethyl alcohol. |
NaOH 40% | 40% (w/v) sodium hydroxide aqueous solution. |
Kovac's Reagent | 5% (w/v) p-dimethylaminobenzaldehyde dissolved in 25% hydrochloric acid and 75% isobutyl alcohol. |
Vaseline Oil | Pure, sterile oil to maintain anaerobic conditions in select wells. |
Test Procedure
- Voges-Proskauer Test (Well 10-VP):
- Add Alpha Naphtol followed by NaOH 40% to the well.
- Mix thoroughly and allow 10–20 minutes for color development.
- Positive Result: Pink/red color.
- Indole Test (Well 11-IND):
- Add Kovac's Reagent to the well.
- Observe for a red/pink layer within a few minutes.
- Positive Result: Red/pink color.
- Anaerobic Maintenance (Wells 2-LDC, 3-ODC, 4-ADC, 7-UR, 8-H2S):
- Seal the wells with Vaseline Oil to create an anaerobic environment.
Refer to the Enterosystem 18R Package Insert for detailed protocols.
Quality Control
- Validate reagent performance using suitable control organisms:
- Positive Controls:
- Escherichia coli: Positive for indole; negative for Voges-Proskauer.
- Enterobacter cloacae: Positive for Voges-Proskauer; negative for indole.
- Negative Controls:
- Strains with no indole or acetoin production.
- Positive Controls:
Storage and Stability
- Store reagents at 2–8°C in their original packaging, protected from light.
- Stability:
- Can tolerate short-term storage or transport at 18–25°C for 4 days or 35–39°C for 48 hours without performance loss.
- Discard reagents if signs of contamination or deterioration are observed.
Precautions
- For in vitro diagnostic use only.
- Classified as hazardous:
- Follow safety protocols as outlined in the Safety Data Sheet (SDS).
- Use by trained personnel with appropriate aseptic techniques.
- Dispose of used reagents and contaminated materials according to laboratory guidelines for biohazard waste.
Applications
- Clinical Diagnostics:
- Identification of Enterobacteriaceae in infections like sepsis, UTIs, and gastroenteritis.
- Industrial Microbiology:
- Quality control for food, water, and pharmaceutical products.
- Research and Education:
- Biochemical profiling of bacterial species.
