Comprehensive Diagnostic System for Veterinary Mycoplasma and Ureaplasma Detection

The Liofilchem® Mycoplasma System VET is an advanced 24-well colorimetric testing system specifically designed for the detection, semi-quantitative enumeration, identification, and susceptibility testing of Mycoplasma spp. and Ureaplasma spp. in veterinary applications. This versatile system enables fast, reliable, and accurate results, making it a valuable tool for veterinary diagnostic laboratories, research facilities, and animal health studies.

    Key Features:

    1. Detection and Enumeration:
      • Simultaneously detect and quantify Ureaplasma spp. and Mycoplasma spp. with a semi-quantitative approach.
      • Indicates colony-forming units (CFUs) through colorimetric changes from yellow to red in wells.
    2. Comprehensive Antibiotic Susceptibility Testing (AST):
      • Includes 9 antibiotics tested at two concentrations for antimicrobial resistance profiling:
        • Tetracycline, Enrofloxacin, Marbofloxacin, Doxycycline, Erythromycin, Clarithromycin, Tylosin, Clindamycin, and Azithromycin.
    3. Multi-Species Applicability:
      • Detects Mycoplasma species across a wide range of animal hosts, including canine, feline, bovine, ovine, caprine, avian, porcine, and reptilian species (see Table 1 in product manual).
    4. Microscopic Detection:
      • Enables visual detection of Trichomonas vaginalis and Candida spp. via microscopic examination of well 6-TR/YE.
    5. Rapid and Reliable Results:
      • Results available in 24-48 hours, allowing for timely intervention and treatment decisions.
    6. Validated Against Reference Strains:
      • Each batch is tested with ATCC reference strains, ensuring quality and reliability:
        • Mycoplasma hominis (ATCC® 23114)
        • Ureaplasma urealyticum (ATCC® 27618)
        • Candida albicans (ATCC® 10231)

    Applications:

    1. Veterinary Diagnostics:
      • Detection and AST for Mycoplasma spp. and Ureaplasma spp. in livestock, pets, and exotic animals.
    2. Research:
      • Ideal for studying antimicrobial resistance and pathogenicity of Mycoplasmataceae in animals.
    3. Environmental and Agricultural Studies:
      • Analysis of avian, bovine, and porcine Mycoplasma in environmental or farm samples.

    Kit Contents:

    • 20 Mycoplasma System VET panels (24-well).
    • 20 Vials of Physiological Solution (7.0 mL each).
    • 1 Instruction Manual, including the Test Results Form.

    Additional Requirements (Not Included):

    • Vaseline oil (ref. 87006).
    • Microscope, cover slides, and slides.
    • Other sundry microbiology laboratory materials.

    Well Configuration and Functionality:

    Detection, Enumeration, and Identification:

    • Wells 1-Uu 103, 2-Uu 104, 3-Uu ≥ 105: Semi-quantitative detection of Ureaplasma spp. (titers from 10³ to ≥10⁵ CFU/mL).
    • Wells 4-Mh 104, 5-Mh ≥ 105: Semi-quantitative detection of Mycoplasma spp. (titers from 10⁴ to ≥10⁵ CFU/mL).

    Microscopic Examination:

    • Well 6-TR/YE: Detection of Trichomonas vaginalis (mobile ciliated trophozoites) and Candida spp. (chlamydospores and hyphae).

    Antimicrobial Susceptibility Testing (AST):

    • Wells 7-24: Testing of 9 antibiotics at two concentrations, with results categorized into sensitive (S), intermediate (I), or resistant (R).
    AntibioticConcentrations Tested (µg/mL)
    Tetracycline4, 8
    Enrofloxacin5, 10
    Marbofloxacin10, 20
    Doxycycline4, 8

    Principle of the Method:

The Mycoplasma System VET works by detecting biochemical changes in the presence of Mycoplasma spp., Ureaplasma spp., or other pathogens. It combines colorimetric reactions and microscopic observations to detect, count, and assess susceptibility to antibiotics.

    How It Works:

    1. Sample Preparation:
      • Swabs or biological fluids are diluted in the provided Physiological Solution for homogeneous distribution.
    2. System Inoculation:
      • Add 0.2 mL of the sample suspension to each well.
      • Add vaseline oil to all wells except 6-TR/YE.
    3. Incubation:
      • Incubate at 36 ± 1°C for 24-48 hours.
    4. Interpret Results:
      • Observe color changes in wells for enumeration and susceptibility testing.
      • Perform microscopic examination for Trichomonas vaginalis and Candida spp..

    Result Interpretation:

    • Positive Reaction:
      • Red or orangey color indicates growth.
    • Negative Reaction:
      • Yellow color indicates no growth.

    Antibiotic Susceptibility Interpretation:

    • Sensitive (S): Growth inhibited at both concentrations.
    • Intermediate (I): Growth inhibited at high concentration only.
    • Resistant (R): No inhibition at either concentration.

    Storage and Stability:

    • Store at 2°C–8°C in the original packaging.
    • Keep away from heat and temperature fluctuations.
    • Shelf life: Valid until the expiry date indicated on the label.

    Validation and Quality Control:

    • Validated with ATCC reference strains, including:
      • Mycoplasma hominis (ATCC® 23114).
      • Ureaplasma urealyticum (ATCC® 27618).
      • Candida albicans (ATCC® 10231).

    Advantages:

    1. Comprehensive Testing: Detects Mycoplasma spp., Ureaplasma spp., Trichomonas vaginalis, and Candida spp.
    2. Antibiotic Profiling: Tests susceptibility to 9 key antibiotics used in veterinary care.
    3. Cost-Effective: Combines detection, enumeration, and AST in a single system.
    4. Easy to Use: Minimal training required for precise results.
