    Procedure:

    1. Preparation of Bacterial Suspension:
      • Isolate the microorganism from an agar plate and suspend in Strepto Broth.
    2. Inoculate System:
      • Transfer 0.2 mL of bacterial suspension into each well.
      • Add PYR disc to well 4-PYR.
      • Overlay well 6-ADC with vaseline oil.
    3. Incubation:
      • Cover the system and incubate at 36 ± 1°C for 18–24 hours.
    4. Interpret Results:
      • Add reagents to specific wells as needed (e.g., VP, IPP, and PYR tests).
      • Record color changes and calculate the 4-digit numerical code using the provided instructions.

    Numerical Code Formation:

    • Tests are grouped into four groups of three:
      • Group 1: VP, IPP, ESC
      • Group 2: PYR, ONPG, ADC
      • Group 3: RIB, ARA, MAN
      • Group 4: SOR, TRE, RAF
    • Assign values (1, 2, or 4) based on positive tests and sum values in each group to form a 4-digit code.
    • Use the code table or online software to identify the microorganism.
