Procedure:

  1. Prepare Bacterial Suspension:
    • Isolate the microorganism on selective agar (e.g., Baird Parker Agar, Vogel Johnson Agar).
    • Suspend colonies in physiological solution to achieve 1-2 McFarland turbidity.
  2. Inoculate the System:
    • Transfer 0.2 mL (4 drops) of bacterial suspension into each well.
    • Add vaseline oil to wells 1-ADC and 2-UR.
    • Place xylose and ribose discs in wells 10-XYL and 15-RIB.
  3. Incubate:
    • Cover the system and incubate at 36 ± 1°C for 18-24 hours.
  4. Interpret Results:
    • Add reagents to specific wells (e.g., VP and NIT tests).
    • Note color changes and use the results to generate a 6-digit code.
    • Identify the microorganism using the code number table.

Storage and Handling:

  • Store at 2°C to 8°C in its original container.
