The Thermo Scientific™ Lambda Exonuclease is a highly processive 5′→3′ exodeoxyribonuclease that specifically digests the 5′-phosphorylated strand of double-stranded DNA (dsDNA). This selectivity makes it a valuable tool for generating single-stranded DNA (ssDNA) from PCR products or cloned DNA fragments.

The enzyme exhibits low activity on ssDNA and non-phosphorylated DNA, ensuring targeted digestion of only phosphorylated strands. It shows no activity at DNA nicks and limited activity at gaps, enabling controlled strand-specific degradation. Lambda Exonuclease is fully active in Thermo Scientific PCR buffers, simplifying integration into molecular biology workflows.
