The Vircell Leptospira IgM ELISA Kit is a high-performance indirect immunoenzyme assay designed for the detection of IgM antibodies against Leptospira interrogans in human serum or plasma. This kit is ideal for diagnosing leptospirosis, a bacterial zoonosis caused by Leptospira interrogans, and provides a user-friendly, efficient solution suitable for manual and automated ELISA systems.

    Key Features

    1. IgM-Specific Detection:
      • Designed to detect IgM antibodies, providing critical information for diagnosing acute leptospirosis infections.
    2. Ready-to-Use Reagents:
      • Pre-prepared reagents reduce handling time and minimize variability.
    3. Simplified Sample Handling:
      • No pre-dilution required; serum/plasma samples can be directly added to the wells.
    4. Automation Compatibility:
      • Suitable for integration into automated ELISA systems, enabling high-throughput testing.
    5. Enhanced Workflow Efficiency:
      • Break-apart wells, color-coded plates, and liquid reagents simplify and optimize workflows.
    6. Stable Reagents:
      • Lyophilized conjugates (when required) ensure long-term stability and consistent performance.

    Principle of the Assay

The Vircell Leptospira IgM ELISA Kit employs an indirect ELISA methodology:

    1. Antigen Binding:
      • Antigens specific to Leptospira interrogans are adsorbed onto the surface of polystyrene wells.
    2. Sample Interaction:
      • IgM antibodies in the sample bind to the adsorbed antigens. Unbound immunoglobulins are washed off.
    3. Conjugate Reaction:
      • An enzyme-labeled anti-human globulin binds to the antigen-antibody complex.
    4. Signal Development:
      • TMB substrate is added, producing a blue-colored solution. Upon addition of the stop solution, the color changes to yellow.
    5. Quantification:
      • The optical density (OD) is measured at 450 nm, and the presence of IgM antibodies is determined based on OD values.

    Applications and Use Cases

    1. Clinical Diagnostics:

    • Acute Leptospirosis Detection:
      • Identify recent Leptospira interrogans infections by detecting IgM antibodies.
    • Serological Screening:
      • Confirm the presence of specific IgM antibodies in suspected leptospirosis cases.

    2. Epidemiological Studies:

    • Seroprevalence Monitoring:
      • Assess the prevalence of Leptospira exposure in populations.
    • Outbreak Investigation:
      • Track and identify sources of leptospirosis outbreaks.

    3. Laboratory Research:

    • Viral Immunology Studies:
      • Explore the immune response to Leptospira infections.
    • Pathogen-Specific Antibody Profiling:
      • Study IgM antibody responses to Leptospira interrogans.
