Liofilchem® Lowenstein-Jensen (LJ) Medium + Pyrazinamide is a ready-to-use, selective culture medium designed for drug susceptibility testing (DST) of Mycobacterium tuberculosis against Pyrazinamide (PZA). Based on the classical egg-based LJ medium, this formulation is enriched with defined concentrations of Pyrazinamide, enabling the detection of resistant strains directly from clinical isolates. This product supports phenotypic drug resistance testing in tuberculosis (TB) control programs, national reference laboratories, and research institutions.

Each tube contains 9 mL of LJ medium supplemented with Pyrazinamide at one of four defined concentrations. The medium supports the slow growth of M. tuberculosis while enabling visual detection of resistance through growth in the presence of the antibiotic.

Key Features

  • Multiple drug concentrations: Enables stepwise or breakpoint-based evaluation of Pyrazinamide resistance.
  • Phenotypic DST for M. tuberculosis: Detects resistance mechanisms not covered by molecular methods.
  • Standardized 9 mL fill volume: Compatible with WHO and CLSI TB DST protocols.
  • Ready to use: Eliminates the need for in-house media preparation and drug supplementation.
  • ISO 9001 & ISO 13485 certified production: Ensures consistent quality and reliability.

Applications and Use Cases

1. Clinical Mycobacteriology

  • Perform Pyrazinamide drug susceptibility testing on:
    • Mycobacterium tuberculosis clinical isolates
    • Suspected MDR-TB and pre-XDR TB cases
  • Supports second-line treatment decisions and TB retreatment regimens.

2. National TB Control Programs

  • Included in phenotypic testing panels for high-priority or drug-resistant TB surveillance.
  • Complements molecular assays (e.g., Xpert, LPAs) by detecting non-genotypic resistance.

3. TB Research and Drug Development

  • Study the activity of Pyrazinamide under different concentrations.
  • Correlate molecular markers (e.g., pncA mutations) with phenotypic outcomes.
  • Use in experimental settings to evaluate resistance stability and mutation frequency.

Use Instructions

  1. Inoculate the LJ + PZA tube with a standardized M. tuberculosis suspension.
  2. Incubate at 37 ± 1 °C for up to 6–8 weeks in a BSL-3 facility.
  3. Examine weekly for colony growth.
  4. Compare growth on PZA-containing tubes to a control tube without drug.
  5. Interpret resistance based on visible growth in the presence of specific PZA concentrations.

Storage and Shelf Life

  • Storage Temperature: 2–8 °C (refrigerated), protected from light.
  • Shelf Life: Up to 6 months from the date of manufacture, when stored unopened.

Liofilchem® Lowenstein-Jensen + Pyrazinamide, 9 mL is an essential tool for phenotypic detection of Pyrazinamide resistance in Mycobacterium tuberculosis. Available in multiple drug concentrations, it enables comprehensive DST profiling in alignment with WHO and national TB program recommendations. Its reliable, ready-to-use format and consistent manufacturing make it an ideal choice for high-complexity TB diagnostic and research laboratories.
