Liofilchem® HEMO-AEROBIC CULTURING is a diagnostic device designed for the isolation and cultivation of fastidious, aerobic, and facultative anaerobic microorganisms from venous blood. The bottles contain Brain Heart Infusion Broth (BHI) enriched with p-Aminobenzoic Acid (PABA) and Sodium-polyanethol-sulphonate (SPS), enhancing the growth of microorganisms while neutralizing inhibitors and preventing blood coagulation.

Product Variants

  1. HEMO-AEROBIC CULTURING (For Adults)
    • Volume: 80 mL per bottle.
  2. HEMO-AEROBIC CULTURING Pediatric (For Children)
    • Volume: 40 mL per bottle.
  3. HEMO-AEROBIC CULTURING Neonatal (For Newborns and children under 2 years)
    • Volume: 9 mL per bottle.

Each variant is tailored to meet specific clinical needs based on patient age and sample volume requirements.

Key Features

  • Nutrient-rich Medium: Brain Heart Infusion Broth supplemented with PABA and SPS supports optimal growth of bloodborne microorganisms.
  • Anticoagulant Properties: SPS prevents blood clotting and neutralizes bactericidal effects of human serum.
  • Wide Microbial Support: Suitable for isolating a wide variety of bacteria, including Gram-positive, Gram-negative, and fungi.
  • Convenient Packaging: Includes bottles with protective caps to ensure sterility and ease of handling.

Medium Composition (g/L)

  • Brain Heart Infusion: 17.0
  • Peptone: 10.0
  • Dextrose: 2.0
  • Sodium Chloride: 5.0
  • Sodium Phosphate Bibasic: 2.5
  • p-Aminobenzoic Acid (PABA): 0.05
  • Sodium-polyanethol-sulphonate (SPS): 0.3
  • Final pH: 7.4 ± 0.2

Clinical Applications

  • Isolation and identification of pathogens in bloodstream infections (BSI).
  • Detection of aerobic and facultative anaerobic microorganisms in cases of septicemia and other infections.
  • Monitoring of bloodstream infections in pediatric and neonatal patients.

How It Works

  1. Sample Collection:
    • Venous blood is drawn aseptically and inoculated into the bottle.
    • Adult samples: 8-10 mL, pediatric samples: 4-5 mL, neonatal samples: 0.5-1 mL.
  2. Inoculation and Incubation:
    • Blood is mixed with the medium to ensure even distribution.
    • Incubate the bottles at 36 ± 1°C for up to 7 days.
  3. Subculturing:
    • Periodically check for microbial growth by subculturing on selective agar plates.
    • Perform Gram staining and microscopic examination to identify microbial morphology.

Advantages

  • Broad Spectrum: Supports the growth of fastidious organisms, including Streptococcus pneumoniae, which may undergo autolysis without appropriate subculturing.
  • Inhibitor Neutralization: PABA neutralizes the effects of sulfonamides, ensuring accurate microbial recovery.
  • Enhanced Anticoagulant Properties: SPS prevents coagulation, minimizes serum bactericidal effects, and partially inactivates certain antibiotics like streptomycin, kanamycin, and gentamicin.

Quality Control

Each batch undergoes quality control using the following reference strains:

  • Escherichia coli ATCC® 25922
  • Staphylococcus aureus ATCC® 25923
  • Klebsiella pneumoniae ATCC® 13883
  • Haemophilus parainfluenzae ATCC® 7901
  • Pseudomonas aeruginosa ATCC® 27853
  • Candida albicans ATCC® 10231
  • And other standard strains.

Limitations

  • Fastidious Microorganisms: Some strains may not grow in the specified incubation period.
  • Inhibitors: Blood samples containing antibiotics may reduce microbial growth.
  • False Negatives: Early disposal of bottles may lead to missed detections.

Safety and Storage

  • Storage Conditions: Store at 10-25°C in original packaging.
  • Shelf Life: Valid until the expiry date on the label.
  • Disposal: Follow laboratory protocols for the decontamination and disposal of biohazardous material.
