Assay Principle
The pre-coated 96-well plate captures hs-CRP from samples. A HRP-conjugated anti-hs-CRP antibody is added, forming a sandwich complex. After incubation and washing, TMB substrate is introduced. A blue color develops, turning yellow upon reaction termination. The optical density (OD) is measured at 450 nm, and the hs-CRP concentration is calculated based on a standard curve.

Sample Types & Preparation
- Serum / Plasma: Collect, centrifuge at 2,000–3,000 rpm for 20 minutes.
- Urine / CSF / Body Fluids: Collect and centrifuge; use supernatant.
- Cell Culture Supernatant: Clarify by centrifugation.
- Tissue Homogenate: Homogenize tissue in PBS (pH 7.4), centrifuge at 4 °C.
Note: Avoid sodium azide (NaN₃), which inhibits HRP. Minimize freeze-thaw cycles. Samples can be stored at -20°C.
Assay Procedure Summary
- Standard Dilution: Prepare six standards (13.5 → 0.75 ng/mL).
- Add 40 μL sample diluent + 10 μL sample to each well (except blank).
- Incubate 30 minutes at 37°C, sealed.
- Wash plate 5× with 1× wash buffer.
- Add 50 μL HRP-conjugated antibody, incubate 30 min.
- Wash, then add 50 μL each of Chromogen A and B.
- Incubate 15 minutes in the dark.
- Add 50 μL stop solution, and read absorbance at 450 nm within 15 minutes.
