Quantitative colorimetric assay for measuring catalase enzyme activity in biological samples
The Invitrogen™ Catalase Colorimetric Activity Kit is a ready-to-use, research-use-only assay designed for the quantification and detection of catalase activity in a variety of biological samples, including serum, plasma, tissues, cells, and erythrocyte lysates. This species-independent assay delivers sensitive, reproducible results using a colorimetric readout at 560 nm.
Catalase plays a critical role in cellular antioxidant defense by decomposing hydrogen peroxide (H₂O₂) into water and oxygen, thereby preventing oxidative damage. The kit enables quantitative measurement of total catalase activity, making it suitable for oxidative stress, antioxidant profiling, and cell physiology research.
Key Features
- High Sensitivity – Detects catalase activity as low as 0.052 U/mL
- Wide Quantification Range – Standard curve range from 0.15–5.0 U/mL
- Broad Sample Compatibility – Works with serum, plasma, tissues, cells, and erythrocyte lysates
- Species Independent – Validated for multiple sample origins
- Complete Kit – Includes all reagents, standards, and 96-well plates for 2 × 96 tests
- Reproducible Results – Optimized reagents ensure high assay precision
Applications
- Measurement of catalase activity in biological fluids and tissue extracts
- Oxidative stress research and antioxidant enzyme profiling
- Studies of hydrogen peroxide metabolism
- Comparative enzyme activity analysis across different species
- Screening of compounds affecting catalase function
Assay Principle
- Sample Preparation – Samples are diluted in assay buffer and added to wells.
- H₂O₂ Reaction – Hydrogen peroxide is added and incubated for 30 minutes at room temperature.
- Colorimetric Detection – Colorimetric Detection Reagent and diluted HRP are added.
- Readout – The HRP reacts with residual hydrogen peroxide to convert the substrate into a pink-colored product measurable at 560 nm. Higher catalase activity results in less residual H₂O₂ and therefore lower absorbance.
Catalase Unit Definition
One unit of catalase decomposes 1 μmol of hydrogen peroxide per minute at 25°C and pH 7.0.
