Principle of Separation

Ficoll-Paque™ PLUS operates on the principle of density gradient centrifugation. When diluted whole blood or buffy coat is carefully layered onto the medium and centrifuged:

• Erythrocytes and granulocytes (higher density) migrate through the medium and pellet at the bottom
• Mononuclear cells (PBMCs) collect at the plasma–Ficoll interface
• Plasma and platelets remain above the gradient

This results in a clean, well-defined mononuclear cell layer suitable for downstream applications.
