It incorporates MUG (4-methylumbelliferyl-ß-D-glucuronide), which is cleaved by β-glucuronidase (GUD), an enzyme specific to E. coli, resulting in a fluorescent end-product that is easily detectable under UV light.

Preparation Instructions
- Suspend 35.7g of dehydrated medium in 1 litre of distilled water.
- Heat gently to dissolve completely.
- Dispense into suitable containers and sterilize by autoclaving at 121°C for 15 minutes.
- Allow the medium to cool before use.
