Thermo Scientific™ Oxoid™ Brilliant Green Agar is a highly selective and differential plating medium designed for the isolation of Salmonella species, excluding Salmonella typhi, from fecal samples, pathological materials, food, and dairy products. Originally described by Kristensen et al. and later refined by Kauffmann, the medium provides effective isolation and differentiation of Salmonella from non-target organisms.
Key Features
1. Selectivity
- Brilliant green inhibits the growth of Gram-positive bacteria and many non-target Gram-negative organisms, making the medium highly selective for Salmonella.
2. Differential Properties
- Contains phenol red as a pH indicator:
- Non-lactose fermenters (e.g., Salmonella): Form red or pink colonies.
- Lactose and sucrose fermenters: Produce yellow colonies with a yellow zone in the medium due to acid production.
3. Broad Applicability
- Suitable for:
- Clinical specimens.
- Food and dairy products.
- Water and environmental samples.
4. Compliance
- Recommended by the APHA and aligns with methods in Standard Methods for the Examination of Water and Wastewater.
Applications
- Clinical Microbiology:
- Detect Salmonella in fecal samples or pathological specimens.
- Food and Beverage Testing:
- Screen dairy products, meat, and processed foods for Salmonella contamination.
- Environmental Monitoring:
- Test water and wastewater for Salmonellae as part of public health surveillance.
- Veterinary Microbiology:
- Detect Salmonella in animal feed and livestock samples.

Preparation Instructions
- Suspend 57.0g of dehydrated medium in 1 litre of distilled water.
- Heat gently while stirring until completely dissolved.
- Sterilize by autoclaving at 121°C for 15 minutes.
- Cool to approximately 50°C, mix thoroughly, and pour into sterile Petri dishes.
Advantages
- Enhanced Selectivity:
- Inhibits the growth of non-target organisms, allowing for reliable isolation of Salmonella.
- Differentiation Capability:
- Distinct color changes help differentiate lactose/sucrose fermenters from non-fermenters.
- Wide Applications:
- Effective across clinical, food, and environmental testing.
- Regulatory Compliance:
- Aligns with USDA and USP standards for microbiological testing.
