The Liofilchem™ ESBL Disc Kit is designed for the confirmation of Extended-Spectrum Beta-Lactamase (ESBL)-producing Enterobacteriaceae. ESBLs hydrolyze most penicillins and cephalosporins, including oxyimino-beta-lactams, but are inhibited by beta-lactamase inhibitors such as clavulanic acid. This kit utilizes the Combination Disc Test (CDT) method to differentiate ESBL-producing isolates from those resistant due to other mechanisms. The test follows CLSI (Clinical and Laboratory Standards Institute) guidelines.

Method Principle
- Combination Disc Test (CDT):
- Compares the inhibition zones of cephalosporins alone (Cefotaxime or Ceftazidime) and in combination with Clavulanic Acid.
- A ≥ 5 mm increase in inhibition zone diameter with clavulanic acid indicates a positive ESBL result.
Test Procedure
- Prepare the Test Organism:
- Use a fresh, pure culture to prepare a suspension equivalent to 0.5 McFarland Standard.
- Inoculate the Agar Plate:
- Spread the suspension evenly over a Mueller Hinton Agar plate using a sterile swab.
- Apply the Discs:
- Place the discs (CTX, CTL, CAZ, and CAL) onto the plate, ensuring adequate spacing.
- Incubation:
- Incubate at 35 ± 2°C for 18-24 hours.
- Interpret Results:
- Measure the inhibition zones and compare as per the following table:
