The Liofilchem® Imipenem+Phenylboronic Acid (IMI+BO) Antimicrobial Susceptibility Discs are specialized diagnostic tools for phenotypic detection of Klebsiella pneumoniae carbapenemase (KPC) and other Class A β-lactamases. These discs combine imipenem (a carbapenem antibiotic) and phenylboronic acid (a boronic acid derivative) to detect resistance mechanisms mediated by serine carbapenemases.

    Key Features

    1. Selective Detection of KPC Activity:
      • Phenylboronic acid inhibits KPC and other Class A carbapenemases, enhancing imipenem activity.
    2. Dual-Disc Synergy Method:
      • Compares zones of inhibition between imipenem-only and imipenem+boronic acid discs to identify KPC-mediated resistance.
    3. Global Standards Compliance:
      • Designed to meet CLSI and EUCAST guidelines for antimicrobial susceptibility testing.
    4. Broad Pathogen Application:
      • Effective for Enterobacterales and non-fermenting Gram-negative bacteria.
    5. Ease of Use:
      • Compatible with routine laboratory workflows using Mueller-Hinton Agar.

    Testing Procedure

    Preparation

    1. Prepare a bacterial suspension equivalent to 0.5 McFarland Standard.
    2. Evenly inoculate the suspension onto a Mueller-Hinton Agar plate.

    Disc Placement

    1. Place the IMI+BO disc alongside an imipenem-only disc (control) with a minimum distance of 25 mm between discs.

    Incubation

    1. Incubate the plates at 35–37°C for 16–20 hours.

    Result Interpretation

    • Measure the zones of inhibition around both discs and compare.

    Result Interpretation

    ObservationInterpretation
Zone for IMI+BO > IMI alone by ≥5 mmPositive for KPC or Class A β-lactamase
Zone diameters nearly identical for both discsNon-KPC carbapenem resistance (e.g., porin loss)
Normal zone diameter for both discsNo carbapenem resistance
