Human Cardiac Troponin T (cTnT) ELISA Kit
The Human Cardiac Troponin T (cTnT) ELISA Kit is a highly sensitive and reliable tool designed for the quantitative detection of cTnT levels in human serum, plasma, urine, cell culture supernatants, and tissue samples. This kit uses the Sandwich-ELISA technique and is ideal for research in cardiac health, myocardial injury, and related conditions.
Purpose
- Quantification of cTnT Levels:
- Measures cTnT concentrations as a key biomarker for cardiac muscle damage.
- Sample Compatibility:
- Valid for human serum, plasma, urine, cell supernatants, and tissue samples.
Principle
The assay employs a Sandwich-ELISA technique to detect and quantify cTnT with high specificity and sensitivity. Below is the detailed process:
- Pre-Coated Plate:
- Microtiter plate wells are pre-coated with an antibody specific to cTnT.
- Sample and Standard Addition:
- Standards and samples are added to the wells, allowing cTnT in the sample to bind to the immobilized antibody.
- HRP-Conjugated Detection:
- A Horseradish Peroxidase (HRP)-conjugated antibody specific to cTnT is added, forming a sandwich complex.
- Washing Step:
- Free and unbound components are washed away to minimize background interference.
- Substrate Reaction:
- TMB substrate solution reacts with HRP, producing a blue color in positive wells.
- Stop Reaction and Measurement:
- A stop solution is added, changing the color from blue to yellow. The optical density (OD) is measured at 450 nm using a spectrophotometer.
- Quantification:
- cTnT concentrations are calculated by comparing OD values to a standard curve.
Sample Preparation
1. Serum Preparation:
- Allow blood to clot at room temperature for 10–20 minutes.
- Centrifuge at 2,000–3,000 rpm for 20 minutes to separate serum.
- Re-centrifuge if precipitates form during storage.
2. Plasma Preparation:
- Collect blood in anticoagulant tubes (e.g., EDTA, citrate).
- Incubate at room temperature for 10–20 minutes.
- Centrifuge at 2,000–3,000 rpm for 20 minutes to obtain plasma.
- Re-centrifuge if necessary.
3. Urine Samples:
- Collect urine in aseptic tubes.
- Centrifuge at 2,000–3,000 rpm for 20 minutes and carefully collect the supernatant.
- Re-centrifuge if precipitates form during storage.
4. Cell Samples:
- For cell secretions, centrifuge culture supernatants at 2,000–3,000 rpm for 20 minutes.
- For intracellular components, dilute cells in PBS (pH 7.2–7.4) and disrupt using freeze-thaw cycles. Centrifuge as above.
5. Tissue Samples:
- Cut, weigh, and freeze tissues in liquid nitrogen; store at -80°C.
- Homogenize tissue in PBS (pH 7.4) at 4°C.
- Centrifuge at 2,000–3,000 rpm for 20 minutes to collect the supernatant.
