    Sample Preparation

    1. Serum Preparation:

    • Allow blood to clot at room temperature for 10–20 minutes.
    • Centrifuge at 2,000–3,000 rpm for 20 minutes to separate serum.

    2. Plasma Preparation:

    • Collect whole blood in tubes with EDTA or citrate anticoagulants.
    • Incubate at room temperature for 10–20 minutes.
    • Centrifuge at 2,000–3,000 rpm for 20 minutes and collect plasma supernatant.
    • Re-centrifuge if precipitates are present.

    3. Urine Samples:

    • Collect urine in aseptic tubes and centrifuge at 2,000–3,000 rpm for 20 minutes.

    4. Cell Samples:

    • For secretions, centrifuge cell culture supernatants at 2,000–3,000 rpm for 20 minutes and collect supernatant.
    • For intracellular components, dilute cells to 1×10⁷/mL in PBS (pH 7.2–7.4), disrupt cells by freeze-thaw cycles, and centrifuge as above.

    5. Tissue Samples:

    • Cut, weigh, and freeze tissue samples in liquid nitrogen. Store at -80°C until use.
    • Homogenize tissue in PBS (pH 7.4) at 4°C.
    • Centrifuge at 2,000–3,000 rpm for 20 minutes and collect the supernatant.

    Important Notes

    1. Timely Processing:
      • Perform sample extraction and ELISA as soon as possible after collection.
      • Avoid repeated freeze-thaw cycles to maintain sample integrity.
    2. Avoid Sodium Azide:
      • NaN₃ inhibits the activity of HRP and is not compatible with this ELISA kit.
